ABSTRACT

Camptothecin and its derivatives are monoterpenoid indole alkaloids exhibiting significant anti-tumor actions. With the aim of improving the production of these pharmaceuticals, the contents of camptothecin and 10-hydroxycamptothecin in different tissues including roots, stems, leaves, young flower buds, opening flowers, fading flowers and seeds from Camptotheca acuminata, were investigated. The young flower buds had the highest alkaloid concentrations (camptothecin, 2.46 mg/g of dry weight; 10-hydroxycamptothecin, 1.41 mg/g of dry weight). Callus showed lower concentrations but it should also be considered as a potential source of these pharmaceuticals. In the present study, the growth rate of Camptotheca acuminata cells in culture did not correlate with contents of camptothecin and 10-hydroxycamptothecin. Alkalo id accumulation by cells under various treatments (heavy metal ions, UV-B), methyl-jasmonate, abscisic acid, salicylic acid and hydrogen peroxide was examined, and the most notable effects appeared in the cells induced by UV-B light (which showed an 11-fold increase in camptothecin concentration) and by salicylic acid (which showed a 25-fold increase in 10-hydroxycamptothecin concentration). These results are significant in the context of the production of both pharmaceuticals.

ABSTRACT

OBJECTIVE@#The merhod of determination of traditional Chinese medicine of aconitum root has been established by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.@*METHODS@#With optimized matrix of 2,5-Dihydroxy- benzoic acid for herbal extract, the identification of aconitum root by MALDI-TOF-MS has been conditioned. By comparison of mass spetra of various of aconitium root, their fingerprint mass-to-charge ratios were found to perform identification. With the characteristic mass-to-charge ratios, It was shown that the mass spectrum of raw aconitum roots were obviously different from that of processed aconitum roots.@*RESULTS@#The raw radix aconiti could be differentiated from radix aconiti preparate by determination of MALDI-TOF-MS.@*CONCLUSION@#The identifical method of aconitum root by MALDI-TOF-MS is rapid, precise and sensitive, it can be used in toxicological analysis of medical tangle and illegal medical practice.

ABSTRACT

Antipyretic activity of methanolic extract of rhizome of N. nucifera was studied on normal body temperature and yeast induced pyrexia in rats. Yeast suspension (10 ml/kg, s.c.) increased rectal temperature after 19 hr of administration. The extract, in doses of 200, 300 or 400 mg/kg (po) produced significant dose dependent lowering of normal body temperature and yeast provoked elevation of body temperature in rats. The effect produced was comparable with the standard antipyretic drug, paracetamol (150 mg/kg, i.p.).

ABSTRACT

Several novel bioactive components isolated from Chinese medicinal plants will be presented. These include novel maytansinoid tumor, inhibitors, some new ent-kaurane and rosane diterpenoids from Mallotus anomalus Meer et Chun (Euphorbiaceae), as well asnovel insecticide, stemona alkaloids from Stemona parviflora C. H. Wright (Stemonaceae). Both are native plants of Hainan island, Chine. 2D NMR techniques such as mono and hetero-COSY, NOESY, COLOC as well as H-NMR line broadening effect were utilized for structure elucidation. The separation techniques, struture elucidations and bioassay results will be reported.
